靶点:HIV Integrase 通路:Anti-infection 背景说明:是一种有效的HIV 整合酶抑制剂。 生物活性:Raltegravir 是一种有效的integrase (IN)抑制剂,作用于WT和S217Q PFV IN,在无细胞试验中IC50分别为90 nM和40 nM。它对HIV-1 IN的选择性比对其他相关Mg2+依赖性酶要高1000倍以上,如HCV聚合酶、HIV反转录酶、HIV RNaseH、人类α-,β-,γ-聚合酶。[1] IC50:Integrase(S217Q PFV): 40nM(IC50);Integrase(WT PFV) : 90nM(IC50) [1] In Vitro:雷特格韦能有效抑制 WT 和 S217Q PFV IN,其 IC50 值分别为 90 和 40 nM。携带 S217H 取代的 PFV IN 对雷特格韦的敏感性降低了 10 倍(IC50 ~ 900 nM)。第二代 INSTI(30)MK2048 对 S217H 内体的 IC50 维持在 200 nM 左右。[1]雷特格韦的代谢方式是葡萄糖醛酸化,而不是肝脏代谢。[2]在镁和钙存在的情况下,雷特格韦的细胞渗透性会降低。[3]雷特格韦和相关的HIV-1整合酶(IN)链转移抑制剂(INSTIs)可有效阻断体外病毒复制并抑制患者的病毒血症。[4]在急性感染的人类淋巴 CD4+ T 细胞系 MT-4 和 CEMx174 中,拉替拉韦(raltegravir)能有效抑制 SIVmac251 的复制。[5] 数据来源文献:[1].
