生物活性：	靶点：Cathepsin;Serine Protease 通路：Metabolic Enzyme/Protease 背景说明：Leupeptin是一种广谱的、具有细胞膜渗透性的蛋白酶抑制剂，能有效抑制丝氨酸、半胱氨酸和苏氨酸蛋白酶。Leupepti还可抑制 Cathepsin B, Cathepsin H 和 Cathepsin L，阻断两性溶酶体的融合。生物活性：Leupeptin， a broad-spectrum covalent inhibitor of serine， cysteine， and threonine proteases， showed inhibitory activity against Mpro， with a 50% inhibitory concentration (IC50) value of 127.2 μM in vitro in our study here. Leupeptin can also inhibit SARS-CoV-2 in Vero cells， with 50% effective concentration (EC50) values of 42.34 μM.[1] In Vitro：Leupeptin， a metabolite produced by plant-symbiotic actinomyces (PSA)， which showed antiviral activity in both Cell Culture and enzyme assays. Western blotting. （1 µM Leupeptin）

MCF-7 cells were treated with the aforementioned CoCl2 concentrations for 24 h and cells were lysed with ice-cold radioimmunoprecipitation assay buffer (150 mM NaCl, 1.0% NP-40, 0.1% Triton X-100, 0.5% sodium deoxycholate, 0.1% SDS and 50 mM Tris-HCl, pH 8.0) and protease inhibitors (AEBSF at 2 mM, Aprotinin at 0.3 µM, Bestatin at 116 µM, E-64 at 14 µM, Leupeptin at 1 µM and EDTA at 1 mM; Beijing Solarbio Science & Technology Co., Ltd., Beijing, China).

Western Blotting（0.5 ug/mL Leupeptin）

The mice heart (n=3) was dissected out and homogenized with a homogenizer at 4 °C in ice-cold lysis buffer containing 150 mM NaCl, 50mM Tris (pH 7.4), 1% sodium deoxycholate, 1%TritonX-100, 0.1% SDS, 25 mM β-glycerophosphate, 2m M sodium pyrophosphate, 1mM EDTA, 1mM Na3VO4 and 0.5 ug/ml leupeptin (Solarbio, Beijing, China). The homogenate was centrifuged at 12,000×g for ten minutes at 4 ℃ to obtain the supernatant which was used to analyze the protein expression.
